تنوع ژنتیکی برخی جمعیت‌هایهندوانه (Citrullus lanatus var. lanatus) بر اساس صفات مورفولوژیک و نشانگرهای ریزماهواره(ISSR)

چکیده

به منظور بررسی تنوع ژنتیکی بین جمعیت‌های هندوانه بومی ایران،24 جمعیت بومی به همراه یک رقم خارجی (دورگ F-1 شرکت ساکاتا) با استفاده از صفات مورفولوژیک و نشانگرهای ISSR مورد مطالعه قرار گرفتند. برای بررسی تنوع مورفولوژیکی، جمعیت‌ها در مزرعه و در قالب طرح بلوک‌های کامل تصادفی در سه تکرار کاشته شده و صفات مورفولوژیک آن‌ها شامل تعداد میوه در بوته، طول بلندترین شاخه، وزن میوه، طول و عرض میوه و ضخامت پوست میوه اندازه‌گیری شد. نتایج تجزیه واریانس نشان داد که بین جمعیت‌‌ها از نظر تمام صفات مورفولوژیک مورد بررسی اختلاف معنی‌دار وجود داشت. گروه‌بندی جمعیت‌ها بر اساس صفات مورفولوژیک با استفاده از تجزیه کلاستر، آن‌ها را در چهار گروه مجزا قرار داد. در بررسی تنوع با استفاده از نشانگر مولکولی، ده آغازگر مورد استفاده 87 مکان قابل امتیازبندی تولید کردند که از این تعداد 75 مکان چندشکل بودند. مقادیرPIC برای آغازگرهای مورد استفاده از 52/0 (آغازگر P4) تا 91/0 (آغازگر UBC864) متغیر بوده و میانگین آن 83/0 برآورد شد. دندروگرام حاصل از تجزیه کلاستر به روش UPGMA مبتنی بر ضرایب تشابه ژنتیکی جاکارد، جمعیت‌ها را در هفت گروه قرار داد. کمترین و بیشترین فاصله ژنتیکی بین جمعیت‌های کردستان و کرمانشاه و جمعیت‌های مازندران و خراسان بود. نتایج این پژوهش نشان داد که نشانگرهای ISSR به طور مؤثری می‌توانند برای مطالعه تنوع ژنتیکی جمعیت‌های هندوانه بومی ایران استفاده شوند و از میان آغازگرهای استفاده شده، UBC864 و UBC823 مناسب‌ترین آغازگرها برای مطالعات بعدی تشخیص داده شدند.

For assessing the genetic diversity of Iranian populations of watermelon, 24 local populations together with one foreign cultivar (F-1 hybrid of Sakata Company) were studied using morphological traits and ISSR markers. Populations were grown in field in a randomized complete block design with three replications, and then morphological traits including fruit number in bush, the tallest banch length, fruit weight, fruit diameter and length and rind thickness were measured. Results of analysis of variance showed significant differences among all populations regarding the morphological traits. Cluster analysis based on morphological traits, divided populations in to four groups. In assessment of genetic diversity by molecular markers, ten used primers generated 87 scorable bands, which 75 sites were polymorphic. Polymorphic information content (PIC) of primers varied from 0.52 (marker P4) to 0.91 (marker UBC864) with anaverage of 0.83. Cluster analysis based on Jaccard genetic similarity coefficient, placed populations into seven main clusters. Minimum and maximum genetic distances were observed between populations of Kordestan and Kermanshah and Mazandaran and Khorasan, respectively. Generally the results of this study revealed that ISSR can be efficiently used for assessment of genetic diversity in watermelon populations native to Iran and UBC823 and UBC864 were recognized as the best primers forfiture studies.
