ژنتیک مقاومت به بیماری سپتوریای برگی گندم در رقم چمران

چکیده

سپتوریای برگی که توسط قارچ Mycosphaerella grminicola ایجاد می‌شود، از بیماری‌های مهم گندم در سراسر دنیا به شمار می‌رود. رقم مقاوم چمران در سطح وسیعی در ایران کشت می‌شود اما ژنتیک مقاومت آن نسبت به این بیماری ناشناخته است. در این تحقیق به منظور بررسی ژنتیک مقاومت رقم چمران نسبت به این بیماری، رقم مذکور با رقم حساس داراب-2 تلاقی داده شد و جمعیت‌های F1، F2، F3 و BC1F2 تهیه شد. نتایج واکنش نتاج حاصل از تلاقی‌های مستقیم و معکوس نسبت به یک جدایه ناپرآزار نشان داد که صفت مقاومت وراثت هسته‌ای داشته و وراثت سیتوپلاسمی در بروز مقاومت نقشی نداشت. در جمعیت هتروزیگوتF1 ، فنوتیپ افراد به صورت نیمه مقاوم و حد واسط ژنوتیپ والدین بود. بررسی‌های فنوتیپی جمعیت‌های F2، F3، و BC1F2 نشان داد که یک ژن با اثر غالبیت غیرکامل، مقاومت رقم چمران به جدایه مورد بررسی را کنترل می‌کند. نتایج آزمایش‌های پرآزاری گذشته و آزمایش‌های مولکولی این تحقیق نیز مشخص کرد که این ژن مقاومت، یک ژن ناشناخته است.

Genetics of Resistance to Septoria Tritici Blotch Disease of Wheat in Cultivar Chamran

Septoria tritici blotch (STB) caused by Mycosphaerella graminicola is one the most important diseases of wheat worldwide. Chamran is a widely cultivated STB-resistant wheat cultivar in Iran but its genetics of resistance to the disease is unknown. To investigate the genetics of resistance of Chamran to STB, the susceptible genotype Darab-2 was crossed with Chamran and F1, F2, F3 and BC1F2 families were prepared. The reaction of progenies derived from direct and reciprocal crosses to an avirulent isolate of the pathogen indicated that the resistance was delivered through nuclear inheritance rather than cytoplasmic inheritance. The heterozygous F1 population displayed a semi-resistant phenotype that was an intermediate between the phenotypes of both homozygous parents. The phenotypic evaluation of F2, F3 and BC1F2 populations indicated that a single incompletely dominant gene in Chamran controls resistance to the studied isolate. The previous results of virulence assays along with molecular experiments performed in this study showed that this resistance gene is an unknown gene.
